Kapa 2G Robust

The question arised, if an engineered enzyme from Kapa Biosystems will work in the presence of DAPI (a fluorescent dye). DNA was extracted from orchids. Several TAQ polymerases were tested before but the results were not satisfactory.
Results:
KAPA Robust together with Buffer B did amplify a product in the presence of 12,50 ng DAPI more info/ in a 25 ul PCR reaction. KAPA Robust and buffer A didn’t result in any bands and the same goes for another enzyme: Advantage 2 from BD Biosciences (Clontech).

683bp product from the TCP1-gene from Listera ovata (an Orchide).
95 °C – 2 min Denaturation
(95 °C – 30 sec, 59 °C – 30 sec, 72 °C – 1:30 min) 30 cycles

KAPA PROBE FAST kits

"We have ran the test KAPA Probe versus ”Other Supplier” on two different machines: ABI 7000 and ABI 7300. It is the same samples that were used all the way (NTC with water also included in the results). Kapa Probe gives a significantly better ”clustering” and therefore a more confident read of the results than the ‘Other supplier’". We have to state that the SNPs used for this experiment are difficult ones!
