GC rich DNA

Dear ,

today we would like to inform you about a new application note from Kapa Biosystems about GC rich DNA. As an example plant genomic DNA is normally difficult to amplify because of an unusual high GC content.

DNA templates with high GC content and short repeats sometimes cause troubles because the formation of secondary structures that resist denaturation makes GC-rich sequences difficult to amplify by PCR.

The application note describes the use of 3 different buffer formulations and the amplification of 10 different amplicons with a GC content from 70 - 83%.

The whole story (including more pictures) is available here: more information/

In comparison to other enzymes in the market, we can state that many suppliers attempt to improve robustness and speed of wild-type polymerases by supplying excessive amounts of enzyme, but it is clear that these overformulated enzymes still do not exhibit the superior performance of KAPA2G Robust Hostart (Data not shown here but available on request).

Before we will go on vacation I would like to show you a nice customer testimonial from Spain about the Kapa Blood PCR kit (thank you very much Javier!) . The customer is the Spanish National Cancer Research Center (http://www.cnio.es/ing/index.asp).

Sample: 2µL fresh and frozen blood and also in parallel with FTA cards (5mm diameter) and followed the KAPA Blood PCR standard protocol.

After a quick check onto an agarose gel, they have sequenced the PCR product.

They were very "surprised" that the kit works so well and we hope we can use this reference lab as a starting point for many other clinical diagnostic labs all over Europe. Here you can find all other customer testimonials about the Kapa Blood PCR kit more

That's all for July, please take care, have a nice summer break and keep in touch if you have any additional questions; requests etc.

Sincerely yours

Juergen
