New buffers for Kapa enzymesDear
Kapa Biosystems will change the buffer for some below mentioned enzymes. Nippon Genetics Europe GmbH will do the replacement of all buffers this week, so if you do not have enzymes on stock, you will automatically receive the new buffers with your orders. Please do not hesittate to contact us if you have any further questions or stocked enzymes.

We are confident that these changes will result in significantly better performance. The buffer changes will affect the follwing products:
KAPA2G Fast: The old buffer A and B will be replaced with the optimized KAPA2G Buffer A. This is a completely different buffer to the old ones, optimized for the unique speed and processivity of the 2G enzyme. Customers should observe improved success rates and amplification specificity with the new, optimized buffer.
KAPA2G Fast HotStart: KAPA2G Buffer A will be the only buffer supplied with this kit and is identical to the Buffer A in the existing kit. The existing Buffer B will be omitted. The existing Buffers A and B were very similar and overall results have shown that they are exchangeable for most applications.
KAPA2G Robust: The non-HotStart kit will now contain the same buffer set developed for the KAPA2G Robust HotStart PCR kit. Buffer A (the same as supplied with KAPA2G Fast and Fast HotStart Kits) is the optimized 2G buffer, in which the old Enhancer 2 has been incorporated. It is therefore not necessary any more to supply the old Enhancer 2 separately. The new KAPA2G Buffer B is similar to the old KAPA2G Robust Buffer A. We have kept this to enable us to supply two very different buffer backbones for optimization. Buffer A again is expected to improve yields and specificity, but Buffer B is generally best for samples contaminated with salts and other inhibitors. The GC Buffer has been optimized for amplification of GC rich amplicons. KAPAEnhancer 1 is still supplied and can be combined with either Buffer A or B.
KAPA2G Robust HotStart: No material changes
KAPAHiFi: The buffers developed for the HotStart kit will now be supplied with the non-HotStart kit. The Fidelity buffer is unchanged, but the GC buffer is improved and now also contains MgCl2. This should make use of the kit easier and improve results with GC rich amplicons
HiFi HotStart: No changes.
In all cases, we expect customers to simply exchange the old buffers for the new ones, without optimization or changes to reaction parameters. Existing KAPA2G Robust users may, however, want to do some evaluation of the new buffers, as these offer a much wider range of options than in the old kit. For such users:
If they used Buffer A or B without any Enhancer before, they can use Buffer B, but should see whether they do not get better results with Buffer A or the GC Buffer.
For applications where Buffer A or B + Enhancer 1 was used in the past, Buffer B + Enhancer 1 is the direct replacement, but Buffer A + Enhancer 1 or the GC Buffer may yield better results.
If they used Buffer A or B with Enhancer 2 in the past, they should use Buffer A now
For all GC-rich applications, first try the GC Buffer. The TDS will also specify other options for GC rich PCR, which include the use of DMSO or Enhancer 1 with the GC Buffer or Buffer A.
