New exciting PCR enzymes - customer referencesDear
We hope you had a nice start into New Year 2009 and hopefully some great days to relax.
Today we would like to focus on customer references for the Kapa Biosystems enzymes. In the last quarter of 2008 we recognized very strong sales for the Kapa SYBR Fast Mixes. The sample to sales ratio is very high (> 70%) because the product is easy to handle and performs much better as all actual competitor kits. Many times, our clients do not need to do any optimization, the kit works on a first trial and outperforms their old mixes. This means you can focus on sales, because there is no need for trouble shooting and reevaluation. Furthermore the QPCR market is showing remarkable growth rates (15% per year) because more and more academic labs need to buy QPCR instruments (for publication).

1) Customer references is the keyword for today. In fact, most of these references are for Kapa 2G Fast HS enzyme. To use a Hot Start enzyme is getting more and more popular because Hot-start technology overcomes the problem of unspecific PCR products. The methodology prevents non-specific extension or degradation of nucleic acid substrates at ambient temperatures by either excluding or reversibly inhibiting the polymerase enzyme. For this reason more and more customers prefer to use a Hot Start enzyme. All Kapa enzymes are blocked by an antibody. Antibody-based hotstart PCR enzymes offer superior performance compared to hotstart formulations based on chemical modification of the enzyme, as enzyme re-activation is faster, more complete and is optimal at pH values which are also optimal for polymerase activity.

A) Beckman Coulter US ( formally Agencourt) switched all of their routine PCR to KAPA 2G Fast Hot Start replacing the Competitor Q Hot Star product, their main motivation was speed and its ability to handle samples with high GC content. D) Johns Hopkins University School of Medicine recently placed a $225,000 order for KAPATaq Antibody Hot Start for the sequencing of one cancer tumour, this replaced Invitrogen’s Platinum Taq which they had been using for several years.
E) Memorial Sloan Kettering Cancer Centres Core Sequencing lab has recently adopted 100% the use of KAPA2G Fast Hot Start for all their PCR work again the motivation was speed and the ability to handle high GC content, they purchase 30,000 per quarter.

3) New product launch - Kapa Long Range HS enzyme
The most of you know that I've been to Japan the last 2 weeks and I realized that the Kapa Long Range enzyme is a wonderful tool to compete against Takara's EXTAQ and LA enzyme. The Long Range enzyme from Kapa is not only applicable for long PCR fragments, it's a very nice tool for difficult PCR reactions leading to high yield and good efficiency. So whenever you run into problems with the above mentioned Takara products you should try this enzyme. The new version is available as an Hot Start enzyme and samples will be available soon.
4) New reaction buffers for some of the Kapa kits
Kapa Biosystems will be consolidating buffers in an effort to align the hot start and non-hot start versions of our products. The differences between the hot start and non-hot start products were a consequence of the product development cycle: the buffer systems would continue to be optimized as the hot start feature was developed. We are now planning to have all the non-hot start products benefit from these buffer improvements and we are confident that these changes will result in significantly better performance and easier kit assembly.
Please note that part codes will be updated to reflect these changes however kit codes will remain the same. Kit assembly documents and technical data sheets will be updated to reflect the changes in part codes and kit format. These changes will take place beginning January 1, 2009. Kapa Biosystems is confident that these changes will provide significant improvements and ease-of-use to the performance of the non-hot start product portfolio. In situations where existing customers are using the non-hot start version of KAPA2G Robust DNA Polymerase Kit, we will provide a simple guide for translating the appropriate buffer and enhancer combinations from one buffer system to the new buffer system. In cases where customers have previously optimized and cannot replicate results with the new buffers we will provide the discontinued buffer system for a limited time to facilitate the transition.
5) Discontinued products:
KAPA2G Fast and KAPA2G Robust non-hot start kits with loading dye will be discontinued. For key accounts Kapa Biosystems will consider supplying buffers with loading dye on a per customer basis
